If we knew what the cell types were in really big datasets with fine scale typing based on other information, then maybe we could define better predictive models to apply to raw gene expression data.

I’m sure connectivity is more defining, but I’d actually be surprised if there isn’t also a reliable transcriptional signature. Since very little rnaseq data comes with connectivity/morphology, analysis is mostly unsupervised cell type discovery.

Rabies has been engineered to act as a retrograde tracer, revealing all the inputs to a neuron. However limitations include toxicity and uncertainty about which synapses it crosses. Furthermore, these viruses are not applicable to all species. 2/11

Just to be clear that the majority of cells in eg Drosophila brain are not *identified neurons* in the sense of just one cell per hemisphere. Instead, they are members of an identified cell type occurring in 2,3,5 or sometimes even several hundred copies per hemisphere.
